生长因子释放体系的研究现状

Research status of growth factor delivery system

摘要: [摘要] 有效的骨再生需要在缺损局部形成持续的成骨刺激。生长因子可以促进骨再生,并动员足量的自身功能细胞迁移到缺损区,适时定向分化,重建牙周组织。但生长因子应用于临床尚存在许多问题,这些问题主要来源于生长因子的使用量和缺乏有效的释放体系。因此,建立有效的时空控制释放体系对于生长因子的合理施用至关重要。目前,常见的生长因子载体主要有以下4种类型:应用胶原膜实现两种生长因子序贯释放;可注射性水凝胶及其衍生构架;不同种类微球的单独和联合应用;纤维支架复合系统。不同的释放系统具有其不同的特点,降解和释放动力学不同,可根据具体的临床需求进行组合和调整。

Abstract: Localized and continuous osteogenic stimulation to defected site is required for effective bone regeneration. Growth factors are very promising molecules to enhance bone regeneration and enable the recruitment of enough endogenous functional cells to the defect regions and promotion of their committed differentiation at appropriate times to reestablish the destroyed tissues. However, the application of the growth factors to clinical practice has been seriously confined, and the problems derived from the determination of the optimal dose of growth factors and the lack of effective delivery systems. Therefore, new delivery systems allowing better control of the spatiotemporal release need to be developed. At present, the growth factor delivery system includes the following four types: sequential delivery using a collagen membrane, injectable polymeric hydrogel and derivative structures, separate and joint application of different kinds of nanoparticles and the composite system of scaffolds. Different delivery systems have their unique characteristics, degradation and release kinetics, and they can be flexibly combined and adjusted according to the clinical needs.
