S100A8在人慢性牙周炎牙龈组织中巨噬细胞表达的研究

Expression of S100A8 on macrophages in gingival tissue with human chronic periodontitis

摘要: [摘要] 目的 观察S100钙结合蛋白A8(S100 Calcium Binding Protein A8,S100A8)在人慢性牙周炎牙龈组织中巨噬细胞(Macrophages, m?)上的表达情况,探讨S100A8在慢性牙周炎发病机制中的作用。方法 将60例受试者牙龈组织标本按牙周炎的病变程度分为三组:正常对照组20例、轻度牙周炎组20例、重度牙周炎组20例。组织标本经4 %?多聚甲醛溶液固定,包埋、切片、 HE染色。各组标本在光学显微镜下观察其组织学变化;经免疫荧光双染色(?double immunofluorescence,DIF),在荧光显微镜下观察S100A8在m?上的表达情况并计算。结果 与正常对照组比较,人慢性牙周炎牙龈组织中CD68-S100A8双阳性m?表达水平均显著升高(P<0.05);重度牙周炎组CD68-S100A8双阳性m?细胞数明显高于轻度牙周炎组(P<0.05)。结论 CD68-S100A8双阳性巨噬细胞与人慢性牙周炎的发生、发展及致病机制有关。

Abstract: Abstract: Objective To observe the expression of S100 Calcium Binding Protein A8 (S100A8) on macrophages (m?) in gingival tissue with chronic periodontitis, to analyze the role of CD68-S100A8 double positive m? in the pathogenesis of chronic periodontitis. Methods The gingival tissue specimens of 60 subjects were divided into three groups according to the degree of periodontitis lesions: normal control group (20 cases), mild periodontitis group (20 cases) and severe periodontitis group (20 cases). Tissue specimens were fixed by 4% paraformaldehyde solution, stained with hematoxylin and eosin for histopathology,stained with double-immunofluorescence for identification of CD68-S100A8 double-positive m? in gingival tissues. Results Compared with the normal control group, the expression of CD68-S100A8 double positive m? in gingival tissues with human chronic periodontitis was significantly increased (P<0.05); the number of CD68-S100A8 double positive m? in severe periodontitis group was significantly higher than that in the slight periodontitis group (P < 0.05). Conclusion The results suggest that the CD68-S100A8 double positive m? may play important roles in the initiation, development and pathogenesis of the human chronic periodontitis.
