利用粪肠球菌感染根管模型评价不同根管封闭剂的抗菌效果

To evaluate the antibacterial ability of different root canal sealers by using the Enterococcus faecalis infected root canal model

摘要: 目的 体外建立粪肠球菌根管感染模型,比较3种生物陶瓷类根管封闭剂的抗菌性能。方法 选择人单直根管的离体前牙48颗,接种粪肠球菌并孵育4周以构建体外根管感染模型。完成根管成形和清理后,随机将样本分为3个实验组和2个对照组,每组按照如下方法进行根管充填:A组,Biodentine+牙胶;B组,iRoot BP+牙胶;C组,MTA+牙胶;D组,阳性对照组(单纯牙胶充填);E组,阴性对照组(无菌根管)。分别于第1、3、7、14天取样,利用涡轮机磨除充填材料和牙本质,收集粉末,梯度稀释后,常规进行细菌培养并计数。结果 3个实验组在4个时间点均表现出对粪肠球菌的抗菌性,与阴性及阳性对照组相比,有统计学意义(P<0.05),3个实验组抗菌效果组间无明显差异(P>0.05)。此外,随时间延长3种材料杀菌效果逐渐增强,第14天达到最高值。结论 Biodentine、iRoot BP、MTA具有相似的抗菌能力,均能有效杀灭粪肠球菌。

Abstract: Objective To establish the model of Enterococcus faecalis root canal infection in vitro and to compare the antibacterial properties of three bio-ceramic root canal sealers. Methods Forty-eight isolated anterior teeth of human single straight root canal were inoculated with Enterococcus faecalis and incubated for 4 weeks to construct an in vitro root canal infection model. After shaping and cleaning, samples were randomly divided into three experimental groups and two control groups. Each group was filled according to the following methods: A, Biodentine; B, iRoot BP; C, MTA; D, positive control group (simple cone filling); E, negative control group (sterile root canal). Samples were taken on day 1, 3, 7 and 14 respectively, and the powder after grinding off the filling material and dentin was collected. After gradient dilution, bacteria were cultured and counted. Results Compared with the positive control group, three experimental groups showed antibacterial activity against Enterococcus faecalis during four time points, which was statistically significant (P<0.05), and there was no significant difference among three experimental groups (P>0.05). In addition, germicidal efficacy of the three materials increased gradually with the extension of time, and reached the highest value in the 14th day. Conclusion Biodentine, iRoot BP and MTA have similar antibacterial properties and can effectively kill Enterococcus faecalis.
