热休克蛋白70对人牙周膜细胞高温损伤影响的研究

Effect of heat shock protein 70 on hyperthermia injury to human periodontal ligament cells

摘要: 目的 观察谷氨酰胺(glutamine,Gln)诱导的热休克蛋白70(heat shock protein,HSP)对人牙周膜细胞高温损伤的影响。 方法 将体外培养的人牙周膜细胞随机分为正常对照组(C组):常规培养的人牙周膜细胞;谷氨酰胺预处理组(Gln组):常规培养的人牙周膜细胞加入Gln 15 mmol/L培养;高温损伤组(SH组):常规培养的人牙周膜细胞置46 ℃培养1 h后恢复常规培养;预处理+高温损伤组(Gln+SH组):常规培养的人牙周膜细胞加入Gln 15 mmol/L培养1 h后,置于46 ℃培养1 h后恢复常规培养。以Western Blot检测细胞内HSP70的表达变化;以四唑盐(MTT)比色法、细胞形态学观察评价细胞存活能力。 结果 Western Blot显示: Gln组、Gln+SH组较C组HSP70表达上调(P<0.05);MTT比色法、细胞形态学观察均显示Gln+SH组较SH组的细胞存活能力强(P<0.05)。 结论 Gln可诱导人牙周膜细胞中的HSP70表达上调,减轻高温对人牙周膜细胞的损伤。

Abstract: Objective To observe the effects of heat shock protein 70 (HSP70) induced by glutamine (Gln) on hyperthermia injury to human periodontal ligament cells.Methods The human periodontal ligament cells cultured in vitro were randomly divided into the following four groups:Control Group (Group C:normal human periodontal ligament cells),Gln Pretreatment Group (Group Gln:normal human periodontal ligament cells were added by Gln 15 mmol/L and cultured for 10 h),Severe Hyperthermia Group (Group SH:normal human periodontal ligament cells were cultured at 46 ℃ for 1 h),and Gln Pretreatment +Severe Hyperthermia Group (Group Gln+SH:after normal human periodontal ligament cells were added into Gln 15 mmol/L and cultured for 1 h,they were cultured at 46 ℃ for another hour).The expression of HSP70 was detected by Western Blot.Cell viability was measured using MTT and cytomorphology.Results Western Blot showed that there was higher expression of HSP70 in Group Gln and Group Gln+SH compared to Group C (P<0.05); MTT and cytomorphology indicated that cell viability was higher in Group Gln+SH than in Group SH (P<0.05).Conclusions Gln can induce higher expression of HSP70 in human periodontal ligament cells,thereby reducing hyperthermia injury.
