p38信号通路在牙周膜细胞成骨分化中的调控作用

The effect of p38 signal pathway on the osteogenic differentiation of periodontal ligament cells

摘要: 目的 研究p38信号通路在人牙周膜细胞成骨分化中的调控作用。 方法 体外培养人牙周膜细胞,取第三代细胞进行成骨诱导培养,实验组加入p38信号通路抑制剂(SB203580),诱导培养4周后通过定量PCR和茜素红染色检测其成骨能力。 结果 成骨诱导可促进牙周膜细胞中p38的磷酸化(p-p38)。抑制p38的磷酸化可降低成骨标记物Runx相关基因(Runx2)、碱性磷酸酶(ALP)、骨桥蛋白(OPN)和骨钙素(OCN)的表达,减少钙结节形成。 结论 p38信号通路在体外培养的牙周膜细胞成骨分化中具有重要的调控作用。

Abstract: Objective To investigate the effect of p38 signal pathway on the osteogenic differentiation of periodontal ligament cells in vitro.Methods Human periodontal ligament cells (PDLCs) was obtained from the third molar and cultured in vitro.Cells from the third passage were cultured in the osteogenic medium, and 10 nM SB203580(an inhibitor of the p38 signaling pathway) was added in the medium in experimental group for 4 weeks.Then the qPCR and Alizarin red S staining were applied to evaluate the osteogenic capability of PDLCs.Results Osteogenic induction could promote the phosphorylation of p38,and inhibition of the phosphorylation of p38 could down-regulate the expression of Runx2,ALP,OPN and OCN,and decrease the formation of calcium nodule,in PDLCs.Conclusions The p38 signal pathway plays a crucial role on the osteogenic differentiation of PDLCs in vitro.
