外周血来源的间充质干细胞与内皮祖细胞体外构建血管化组织的研究

Study of vascularization in vitro with peripheral blood-derived mesenchymal stem cells and endothelial progenitor cells

摘要: 目的 利用两种外周血细胞在体外构建出血管样组织。方法 从外周血提取内皮祖细胞与间充质干细胞并鉴定,在纤维蛋白凝胶中以不同的密度培养,在显微镜下观察至第7天。结果 兔外周血可以提取出内皮祖细胞与间充质干细胞,内皮祖细胞与间充质干细胞的比值为100万/mL:200万/mL时可以在体外形成血管样结构。结论 外周血中可以提取出特定细胞并在体外共培养形成血管样组织。

Abstract: Objective To engineer vascularized tissue with two types of cells from peripheral blood. Methods Endothelial progenitor cells (EPC) and peripheral blood-derived mesenchymal stem cells (PBMSC) were isolated from rabbit peripheral blood and identified. Then, they were cultured in fibrin at different ratios under the observation of a microscope until the 7th day after the initial creation. Results EPC and PBMSC were successfully isolated from rabbit peripheral blood. Vascularized tissue were acquired at a certain ratio (EPC were 1 million cells/mL, PBMSC were 2 million cells/mL). Conclusion Certain cells isolated from peripheral blood can be used to acquire vascularized tissue by co-cultured strategy at a certain density in vitro.
