Effect of Hydroxycamptothecin on proliferation,migration,cellcycle distribution and apoptosis in human pharyngeal squamous carcinoma cell line FaDu

摘要: 目的 探讨羟基喜树碱对人咽鳞癌细胞系FaDu增殖、迁移、凋亡及周期的影响。方法 培养人咽鳞癌细胞系FaDu,采用MTT实验、Transwell 细胞迁移实验和流式细胞仪检测不同浓度的羟基喜树碱对FaDu细胞增殖、迁移、凋亡及周期的影响。结果 MTT实验、Transwell侵袭实验结果表明一定浓度羟基喜树碱可以抑制Fadu细胞的增殖和迁移能力,流式细胞仪检测结果显示80ng/ml羟基喜树碱可以将FaDu细胞阻滞在S期并促进其凋亡。结论 羟基喜树碱可以抑制FaDu细胞的增殖和迁移能力并诱导其凋亡,且羟基喜树碱对FaDu的凋亡诱导作用与其对FaDu细胞周期的影响有关。

Abstract: Objective To investigate the effect of Hydroxycamptothecin on the proliferation,apoptosis,migration and distribution of the cellcycle in FaDu after exposed to HCPT. Methods Human pharyngeal squamous carcinoma cell line FaDu was cultivated. The growth inhibition was evaluated by MTT assay and the migration was measured by Transwell migration assay. The apoptosis and the cell cycle distribution were detected by flow cytometry after the treatment with HCPT. Results MTT and Transwell experiments showed that certain concentration of HCPT could effectively inhibit the growth and migration of cell line FaDu. According to the results of flow cytometry,the cell cycle was arrested in S phase and apoptosis was obviously induced after treated with HCPT(80ng/ml).Conclusions HCPT can inhibit the growth and migration of cell line FaDu and induce its apoptosis. HCPT induced apoptosis of the cell cycle in FaDu is probably connected with its effect on the cell cycle of FaDu.
