SMAD4在舌鳞状细胞癌中的表达及意义

Expression and significance of SMAD4 in tongue squamous cell carcinoma

摘要: 目的 检测SMAD4在舌鳞状细胞癌中的表达水平,并分析其与肿瘤细胞分化程度及淋巴结转移之间的相关性。方法 收集正常口腔黏膜组织石蜡标本8例、舌鳞癌组织石蜡标本38例。采用免疫组化方法检测正常组织及肿瘤组织中SMAD4表达水平。结果 SMAD4蛋白在舌鳞癌组织中的表达显著低于正常舌黏膜组织(P<0.05)。SMAD4蛋白在癌细胞中的整体表达水平在高分化鳞癌与中低分化鳞癌组中无明显统计学差异(P>0.05),但其核表达量在高分化鳞癌组显著高于中低分化组(P<0.05)。伴淋巴结转移鳞癌组中SMAD4在细胞核内的表达量低于不伴淋巴结转移鳞癌组(P<0.05)。结论 SMAD4在舌鳞状细胞癌中的表达水平,尤其是核表达水平与舌鳞癌的细胞分化程度及淋巴结转移相关。

Abstract: Objective To detect the expression levels of SMAD4 in Tongue Squamous Cell Carcinoma (TSCC) and analyze its correlation with cellular differentiation degree and lymph node metastasis. Methods 8 cases of normal oral mucosa paraffin specimens, 38 cases of TSCC paraffin specimens were collected in the present experiment. Immunohistochemical staining was used to detect the expression levels of SMAD4 in normal and TSCC tissues. Results SMAD4 protein in squamous cell carcinoma tissues showed lower expression level than in normal tongue mucosa tissues(P <0.05). Well differentiated TSCC and moderately or poorly differentiated TSCC had no significant difference in overall expression levels of SMAD4 protein(P>0.05). But the SMAD4 expression in nuclei of the cells in well differentiated TSCC group was higher than that in moderately or poorly differentiated TSCC group(P<0.05). SMAD4 expression in nuclei of the cells in non- metastatic TSCC group was higher than that in metastatic TSCC, especially in moderately or poorly differentiated TSCC group (P <0.05). Conclusion The expression level of SMAD4 in TSCC , especially in nuclei of the cells correlates with differentiation levels of TSCC cells and tumor metastasis.
