不同浓度的抗菌肽RISE-AP12?对体外培养成骨细胞生物活性的影响

Effects of different concentrations of antimicrobial peptide RISE - AP12 ? on the bioactivity of cultured osteoblasts in vitro

摘要: 目的 :通过检测不同浓度的抗菌肽对成骨细胞的增值、迁移、成骨、蛋白合成、碱性磷酸酶活性、细胞传代的影响,探讨抗菌肽局部给药的效应浓度范围。方法 :将MG63细胞预培养24h,将不同浓度的抗菌肽(0、5 、10、15、20、25、30ug/ml)加入MG63成骨细胞培养液中,继续孵育1d、2d、3d、4d 、21d后,倒置显微镜观察细胞的形态、(1、2、3d)后用划痕法检测细胞的迁移能力、并与2d后检测其对细胞的增值活性及蛋白合成的影响,4d后检测其对细胞碱性磷酸酶活性的影响、21d后茜素红染色检测各组成骨钙结节的形成;细胞传代培养3次,MTT检测成骨细胞增值活性,显微镜观察成骨细胞形态。结果: 0-20ug/ml抗菌肽对成骨细胞的形态无影响,对细胞的增值和蛋白合成及碱性磷酸酶活性没有影响,对细胞的传代培养没有影响;25-30ug/ml抗菌肽对成骨细胞的形态有影响,抑制细胞的增值和蛋白合成,促进碱性磷酸酶的活性,对细胞的传代有影响。结论:在抗菌肽抑菌、杀菌的效果范围内,10-20ug/ml抗菌肽可作为抗菌肽局部给药的效应浓度范围。

Abstract: objective: by detecting different concentrations of antibacterial peptide of osteoblast value-added, migration, osteogenesis, protein synthesis, the influence of alkaline phosphatase activity, cell batches, explore antibacterial peptide effects of the local drug concentration range.Methods: 24 h MG63 cells culture, the different concentrations of antimicrobial peptide (0, 5, 10, 15, 20, 25, 30 ug/ml) to join MG63 osteoblast culture, continue to incubation 1 d and 2 d, 3 d, 4 d and 21 d, inverted microscope to observe the cell morphology, (1, 2, 3 d) after the scratch method to detect cell migration ability, and with 2 d after testing its value-added activity of cells and protein synthesis, the effect of 4 d after testing its effect on the cell activity of alkaline phosphatase, alizarin red staining test groups of osteogenesis after 21 d calcium nodule formation;Cell subculture 3 times, determined by MTT detection osteoblast value-added activity, microscope observation osteoblast form.Results: 0-20 ug/ml of antimicrobial peptide form of osteoblast, on cell proliferation and protein synthesis and alkaline phosphatase activity has no effect, has no effect on cell subculture;25 to 30 ug/ml of antimicrobial peptides have an effect on the morphology of osteoblast, inhibition of cell proliferation and protein synthesis, promote the activity of alkaline phosphatase, batches of cells.Conclusion: the effect of antibacterial peptides in antibacterial, sterilization, 10-20 ug/ml antibacterial peptides can be used as antibacterial peptide effects of the local drug concentration range.
