高通量测序方法分析实验室培养对人体口腔唾液菌群的影响

Effect of culture in vitro on human salivary microflora by high throughput sequencing

摘要: 目的 研究实验室培养对口腔唾液菌群结构及其多样性的影响。方法 本实验选取6个健康且符合纳入排除标准的志愿者,采集口腔唾液菌群样本并进行实验室培养。基于Illumina Hiseq测序平台,我们利用16S rDNA高通量测序技术,研究实验室培养对人体口腔唾液细菌的影响,分析了培养前后的菌群物种丰度和菌群结构差异。结果 高通量测序一共得到618个OTUs,测序分析显示人体口腔唾液菌群在实验室培养前后,菌群生物多样性的差异无统计学意义(P>0.05),菌群构成的差异无统计学意义(P>0.05)。OTUs Venn图显示实验室培养前后,人体口腔唾液菌群拥有大约78.4%相同物种。结论 16S rDNA高通量测序分析显示,人体口腔唾液菌群在实验室培养前后具有相似性。

Abstract: Objective To study the effect of culture in vitro on oral microfloral structure and biodiversity. Methods Six healthy and eligible volunteers were selected. Oral bacterial samples were collected and cultured in vitro. Based on Illumina Hiseq sequencing platform, 16S rDNA high throughput sequencing technology was used to study the effect of culture in vitro on human oral microflora, and then the structure and relative abundance of microflora before and after culture were analyzed. Results There were a total of 618 OTUs in this sequencing. Alpha diversity showed that the difference of biodiversity had no statistical significance before and after culture in vitro. Beta diversity showed that no significant difference was observed in bacterial structure. According to the OTUs Venn diagram, there were about 78.4% identical OTUs before and after culture in human oral salivary flora. Conclusion 16S rDNA high throughput sequencing analysis shows that human oral salivary flora has similarity before and after culture in vitro.
