肝细胞生长因子对成骨细胞增殖、凋亡及成骨分化的影响

Effects of hepatocyte growth factor on proliferation, apoptosis and osteogenic differentiation of mouse osteoblasts

摘要: [摘要] 目的 探讨肝细胞生长因子(hepatocyte growth factor,HGF)对小鼠成骨细胞(MC3T3-E1)的增殖、凋亡和成骨分化的影响。方法 选择细胞计数试剂盒和TUNEL法检测10 ng/mL和25 ng/mL的HGF对MC3T3-E1细胞增殖和凋亡的影响。设置对照组和实验组(25 ng/mL),应用碱性磷酸酶和茜素红染色检测细胞的矿化能力;应用RT?PCR和Western Blot检测成骨相关因子的mRNA及蛋白的表达水平。结果 25 ng/mL的HGF在48 h和72 h时均可促进细胞增殖,在24 h、48 h和72 h时均可抑制细胞凋亡。实验组钙结节大小和数量明显高于对照组,矿化程度显著增高。实验组细胞的成骨相关因子ALP、OCN、Runx2、OSX的mRNA和蛋白表达水平均高于对照组。结论 HGF能够促进MC3T3-E1细胞的增殖和成骨分化,并抑制其凋亡。

Abstract: [Abstract] Objective To investigate the effects of hepatocyte growth factor (HGF) on proliferation, apoptosis and osteogenic differentiation of mouse osteoblasts (MC3T3-E1). Methods Cell counting kit-8 and TUNEL method were used to detect the effects of HGF on proliferation and apoptosis of MC3T3-E1 cells at different concentrations (10 ng/mL and 25 ng/mL). The control group and the experimental group (25 ng/mL) were set and the mineralization capacity of the cells was detected by alkaline phosphatase and alizarin red staining. The mRNA and protein expression levels of osteogenesis related factors were detected by RT-PCR and Western blot. Results HGF at a concentration of 25 ng/mL promoted cell proliferation at 48 h and 72 h, and inhibited apoptosis at 24 h, 48 h and 72 h. The size and number of calcium nodules in the experimental group were significantly higher than that in the control group which means the degree of mineralization was significantly increased. The mRNA and protein expression levels of osteogenesis related factors including ALP, OCN, Runx2 and OSX in the experimental group were significantly increased than those in the control group. Conclusions HGF can promote the proliferation and the osteogenic differentiation of MC3T3-E1 cells and inhibited their apoptosis.
