Effects of IL-17 on the expression of RANKL of human periodontal ligament cells

Abstract: Objective To investigate the effect of interleukin-17(IL-17) on the expression of RANKL and to explore the possible pathogenic mechanism of IL-17 regulating periodontitis bone reconstruction. Methods hPDLCs were treated with IL-17 of variable concentrations (0、10、25、50 ng/mL),and then the optimal concentration was used in the time-dependent experiment. The expressions of RANKL were assayed by real-time RT-PCR and western-blot. Results The expressions of RANKL were significantly up-regulated by IL-17 in a dose-dependent and time-dependent manner. Conclusions IL-17 can up-regulate the expression of RANKL of hPDLCs,suggesting that IL-17 can modulate the alveolar remodeling via regulating the expressions of pro-osteoclastic molecules of hPDLCs.
