摘要: N6-甲基腺嘌呤(N6-methyladenosine,m6A)是真核生物信使RNA中最普遍的一种修饰。Wilms肿瘤蛋白1相关蛋白(Wilms'tumor 1-associating protein,WTAP)作为m6A甲基化转移酶的调控亚基,与 甲基转移样蛋白3(methyltransferase-like protein 3,METTL3)、甲基转移样蛋白14 (methyltransferase-like protein 14,METTL14)组成WMM复合物,催化m6A的产生。大量证据表明,WTAP可以通过调控RNA代谢,影响肿瘤的发生和进展,并与肿瘤的预后有着显著的相关性。本篇综述将关注WTAP的生物学功能和肿瘤中的作用,以及其作为肿瘤治疗潜在靶点的可行性。

Abstract: N6-methyladenosine is considered the most common transcript modification in eukaryotic messenger RNA. WTAP (Wilms' tumor 1-associating protein), as a regulatory subunit of m6A methyltransferases, combines with METTL3 and METTL14 into WMM complex, which catalyzes m6A. Accumulating evidence suggests that WTAP plays an important role in tumorigenesis and cancer progression by regulating RNA metabolism, and has a significant association with prognosis of several kinds of cancer. This review will focus on the biological function of WTAP and its role in cancer, as well as its feasibility as a potential target for tumor therapy.
