MicroRNA27b inhibits cell proliferation of oral squamous cell carcinoma through targeting FZD7

Abstract: Objective To investigate the role of microRNA27b (miR27b) in regulating the cell proliferation of oral squamous cell carcinoma and the underlying molecular mechanism. Methods Cell proliferation was measured by MTT assay. The expression of miR27b, the mRNA expression levels of Frizzled7 (FZD7), cyclin D1 and cmyc were detected by realtime quantitative polymerase chain reaction (qPCR). The protein expression of FZD7 was detected by Western blot analysis. The targeted relationship between miR27b and FZD7, and the activity of Wnt signaling pathway were examined by luciferase reporter assay. Results The expression of miR27b in oral squamous cell carcinoma cell lines was significantly decreased. Overexpression of miR27b markedly inhibited the cell proliferation of oral squamous cell carcinoma. miR27b could target and inhibit FZD7 expression and suppress Wnt signaling pathway. Conclusions miR27b could inhibit the cell proliferation of oral squamous cell carcinoma through targeting and inhibiting FZD7 and FZD7mediated Wnt signaling pathway.
