Construction of a specific biomineralization template of enamel

Objective Construct a self-assembly amelogenin-inspired oligopeptide amphiphile as organic matrix template for simulating mineralization of enamel. Methods Inspired by amelogenin that self-assembles into nanospherical supermolecular structures in enamels and according to the design features of "self-assembly polypeptide biological materials", a novel biomineralization template of "amelogenin-like oligopeptide amphiphile" was designed. The oligopeptide amphiphile was synthesized by standard solid-phase peptide synthesis, purified and characterized by high performance liquid chromatography (HPLC) and mass spectrometry (MS). Peptide self-assembly was induced with 1 M CaCl2 solution and characterized with scanning electron microscope (SEM) and transmission electron microscope (TEM). Ca2+-induced self-assembled peptides were smeared onto a holey carbon-coated TEM grid and fixed with 2.5% glutaraldehyde at room temperature for 1 h. Grids loaded with self-assembled peptide were immersed in 10 mM CaCl2 solution for one hour at room temperature or 40 oC in oven and then rinsed in deionized water. The samples were then exposed to 5 mM Na2HPO4 solution as did in CaCl2 solution followed by rinsing. This cycle was repeated five times, after which the specimens were removed, washed with deionized water, dried in atmospheric conditions, and examined with SEM, TEM and energy dispersive spectrometer (EDS). Results The sequence of the amelogenin-inspired oligopeptide amphiphile was C18H35O-Thr-Lys-Arg-Glu-Glu-Val-Asp. Peptide molecule weight (MV) and purity were 1142.41 and 98.29% accordingly, identified with MS and HPLC respectively. The oligopeptide amphiphile could self-assemble into a gel by Ca2+ induction. TEM showed that the self-assembly hydrogel consisted a β-sheet conformation nanofiber networks; the peptide fiber could adsorb calcium and phosphate ions from the solution. Then, minerals crystals nucleated, grew on the surfaces of the peptide fibers, and orientated along the fibers. Conclusions A novel amelogenin-inspired oligopeptide amphiphile was designed and synthesized, which could be a specific template for biomimetic mineralization of enamel, and could be useful in the study of inducing enamels' remineralization .
Analysis of correlation between IL-6, L-6R and IL-4 single nucleotide polymorphism and the susceptibility of chronic periodontitis among Shanghai patients of Han nationality

Objective To investigate the correlation between IL-6、IL-6R and IL-4 single nucleotide polymorphisms (SNPs) and the susceptibility of chronic periodontitis among Shanghai patients of Han nationality. Methods Through the application of TaqMan probe real-time fluorescent quantitative PCR, 128 health control subjects and 198 patients with chronic periodontitis(CP) above moderate degree were investigated from three single nucleotide polymorphisms sites, namely IL-6 -572G/C,IL-6R 48892A/C and IL-4 -590C/T. Statistical analysis of Chi square test was performed with SPSS17.0 software package. Results In the screening of the 3 target SNPs,there were no statistically significant differences in distributions of the genotypes and alleles between healthy controls and CP patients. Conclusion The result suggests that the SNPs gene locus of IL-6 -572G/C, IL-6R 48892A/C and IL-4 -590C/T may not be significantly associated with the susceptibility of chronic periodontitis among Shanghai patients of Han nationality.
Expression of proliferating cell nuclear antigen in periodontal ligament during orthodontic tooth movement

Objective  To investigate the changes of proliferating cell nuclear antigen(PCNA) in rats' periodontal ligament in orthodontic tooth movement and the mechanism of changes of PCNA during orthodontic tooth movement. Methods Young SD rats were used to build up orthodontic movement models. The animals were sacrificed 24 hours,3 days,5 days,7 days,14 days and 21 days after continuous mechanical stress was applied. The expressions of PCNA were performed by immunohistochemistry. Results Expression of PCNA in rats' periodontal ligament was weak in the control group. In the experimental group, the expression of PCNA on the tension side of periodontal ligament got stronger 24 hours after continuous mechanical stress, strongest 3 and 5 days after continuous mechanical stress, weaker 7 days after the stress, and normal 21 days after the stress. Conclusion  Orthodontic treatment promotes cell proliferation in periodontium, and completes reconstruction of periodontium.
Application of GuttaFlow in C-shaped canals in mandibular second molars

Objective To evaluate the efficacy of GuttaFlow in C-shaped canals in mandibular second molars. Methods Select 80 teeth in 76 cases of C-shaped canals in mandibular second molars that needed treatment. The teeth were randomly divided into two groups, 40 in each. Root canals in the experimental group were filled with GuttaFlow and gutta-percha points. Root canals in the control group were filled with cortisomol and dealed with cold lateral condensation of gutta-percha. Compare the filling time, quality of root canal filling according to X-ray photos, pain within one week after the treatment, and curative effects one year after the treatment. Results The filling time of experimental group was obviously shorter than that of the control group. The occurrence of postoperative pain was low. The quality of root canal filling and curative effects were better than those of the control group. The difference was significant between two groups(P<0.05). Conclusion GuttaFlow is easy to operate in C-shaped canals in mandibular second molars. It's effective and worth popularization and application in clinics

Abstract: Objective Compare remediation effectiveness of traditional full denture with that of magnetic attachment full denture. Method From May, 2008 to October, 2010, 17 patients seeking for full denture remediation in Taizhou Hospital of Chinese medicine were chosen to wear traditional full denture for three months and then to wear magnetic attachment full denture for three months. Investigate the patients’ satisfaction by questionnaire survey and detect the masticatory efficiency with weighing method 3 months after treatment. Result The patients were more satisfied and have higher masticatory efficiency with magnetic attachment full denture compared with traditional full denture. Conclusion Magnetic attachment full denture is obviously better than traditional full denture in terms of fixity, comfortableness and masticatory efficiency.
Influence of periodental non-surgical treatment on OC in gingival crevicular fluid of patients with aggressive periodontitis

Objective To investigate the influence of periodontal non-surgical treatment on periodontal clinical parameters and OC in gingival crevicular fluid(GCF) of patients with aggressive periodontitis. Experiments To detect periodontal clinical parameters of 60 teeth from 20 cases of aggressive periodontitis before treatment and 4 weeks after periodontal non-surgical treatment. Assays of OC in GCF were performed by ECL. Results Level of OC was distinctly higher in experimental group than that in the control group; level of OC obviously reduced after teatment(p<0.05). Conclusion Periodontal non-surgical treatment can distinctly improve the clinical parameters and decrease the amount of OC in GCF of patients with aggressive periodontitis.

Research on dental health care awareness and situation among pregnant women in Yancheng

Objective To study the level of knowledge on dental care during pregnant and puerperal period among gravidas in Yancheng. Methods We performed a survey covering 16 questions concerning dental care knowledge among 1206 gravidas. A routine dental inspection was conducted for all surveyed subjects. Statistical analysis was done with factors as ages and gestational weeks and with data acquired from the survey. Results Local gravidas were generally in lack of dental care knowledge, given that 91.34% of gravidas had not received any dental care education; dental health levels were low given that 56.30% of them suffered from saprodontia and 75.04% from gingivitis; 73.63% of them had soft deposits and 51.33% had dental calculus. However, the proportions of gravidas with saprodontia and gingivitis seeking for medical care were only 1.37% and 5.66%. Conclusions Dental health education and practice improvement are suggested for local gravidas.
Survey on oral health of 5 352 middle school students in Wuhu

Objective To understand the oral health conditions of middle school students in Wuhu in order to explore effective measures to prevent and reduce oral diseases. Methods 5352 students in middle schools who underwent physical examination in our hospital in December, 2011 were investigated in this study according to the criterion issued by World Health Organization(WHO)on October 2011.Statistical analysis was done. Results Among the 5352 middle school students in Wuhu,prevalence rate of caries and gingivitis was 38.5% and 19.1%, index of fluorosis was 0.0197, malocclusion rate was 92.9%. Statistical analysis showed that rural students had more serious caries than urban students and that girls had more serious caries than boys. There was statistical significance. Analysis showed that urban students had more gingivitis than rural students, so did the boys than girls. There was also statistical significance. Incidence rates of malocclusions did not show significant difference between boys and girls. Conclusion Caries prevalence rate of middle school students in Wuhu was similar to that of the country. There oral health status was better than the national status. Malocclusion was in high incidence. Rate of visiting Department of stomatology was low.
