摘要:

溶酶体是维持牙周组织稳态的重要细胞器,其功能障碍在牙周炎的发生发展中发挥关键作用。牙周炎中持续的酸性微环境、病原刺激及过度炎症反应,可引起溶酶体酸化、自噬流及膜稳定性等多层面功能失衡,进而影响病原体清除、免疫调控和骨代谢稳态,推动炎症反应级联放大和牙槽骨破坏。本文旨在综述溶酶体及其功能障碍对牙周炎影响的研究进展,以期为深入理解该疾病的分子机制及药物治疗提供新的视角和理论依据。

Abstract:

Lysosomes are essential organelles for maintaining periodontal tissue homeostasis, and their dysfunction plays a critical role in the initiation and progression of periodontitis. In periodontitis, a persistently acidic microenvironment, continuous pathogenic stimulation, and excessive inflammatory responses can induce multi-level lysosomal dysfunction, including impaired lysosomal acidification, disrupted autophagic flux, and compromised membrane stability. These alterations subsequently affect pathogen clearance, immune regulation, and bone metabolic homeostasis, thereby promoting inflammatory cascade amplification and alveolar bone destruction. This review aims to summarize recent advances in research on the roles of lysosomes and their dysfunction in periodontitis, with the goal of providing new perspectives and a theoretical basis for a deeper understanding of the molecular mechanisms and pharmacological interventions of this disease.
